Tobacco has been one of the most frequently used materials for plant
tissue culture in many aspects of research such as callus culture cell
suspension culture, single cell culture, differentiation mechanisms, and
anther culture for haploid plants Accordingly, many investigations have
been carried out on the chemical composition of the culture medium and the
media have been changed from natural nutrients to synthetic ones, although
they were mostly devised for agar culture. One of the most frequently used
media for plant callus is that presented by Linsmaier and Skoog, generally
called RM-1964 medium. It is also preferable for the growth of tobacco
cells in suspension culture. We usually add 2,4-dichlorophenoxyacetic acid
(2,4-D) at 0.2 mg/l, thiamine hydrochloride at 0.4 mg/l, and sucrose at 30
g/l to the RM-1964 inorganic medium. In order to obtain basic information
for a large scale tobacco cell suspension culture, some investigations on
the tobacco species suitable for the culture, nutritional conditions, and
chemical constituents of the cultured cells, etc., have been undertaken.
