Title Establishment of a cell line of extrahepatic bile duct carcinoma.

Abstract To investigate the cellular biological charateristics of bile duct carcinoma, the first cell line of extrahepatic bile duct carcinoma, named as QBC939, was established in China. The cell line was histologically similar to the originai tumor. Electron microscopic studies indicated the presence of characteristic junctions between cells, microvilli on the cell surface, microfilaments and microtube in cytoplasm. Immunohistochemistry of cultured cells and reconstituted tumor in nude mouse showed that the cell line was strongly positive for karatin and desmosome, supporting the epilhelial origin of the cell line. The study of growth kinetics demonstrated that the QBC939 cell line grew fast with independent and higher viability. The cytogenetic study revealed that the abnormality in number and structure, such as fracture, transplantation, loss and insert of chromosomes, presented in the cell line. Soft sepharose culture and nude mouse xenografts indicated that this cell line was strungly tumorigenic.
