Cellosaurus FIRE2FLIPCS clone 1 (CVCL_E6HT)

[Text version]
Cell line name FIRE2FLIPCS clone 1
Synonyms FIRE2FLPCS-520-8
Accession CVCL_E6HT
Resource Identification Initiative To cite this cell line use: FIRE2FLIPCS clone 1 (RRID:CVCL_E6HT)
Comments Characteristics: Expresses a Fe-IRE (FIRE) biosensor that senses IRP activity and cellular iron status (PubMed=39824193).
Characteristics: This cell line contains a FIRE2 biosensor (emiRFP670-sensor, mRuby3-control) and is transposed with two PiggyBac constructs FIRES-FLIP-PBCS-E670 and FIREC-PBCS-MR3 flanking FIRE sensor and control fluorescent protein, respectively. The FLIP-FIRE sensor is composed of a 740 bp sequence synthesized in backwards orientation and based on known mammalian IRP1/2 binding sites (PubMed=39824193).
Genetic integration: Method=PiggyBac transposition; Gene=FPbase; YHJV5; emiRFP670 (Note=Constitutively fluorescent near IR fluoresent protein from R.palustris).
Genetic integration: Method=PiggyBac transposition; Gene=FPbase; MZMFB; mRuby3 (Note=Bright red fluorescent protein derived from eqFP611).
Derived from site: In situ; Blastocyst; UBERON=UBERON_0000358.
Cell type: Embryonic stem cell; CL=CL_0002322.
Species of origin Mus musculus (Mouse) (NCBI Taxonomy: 10090)
Breed/subspecies: C57BL/6J x 129S4/SvJae.
Hierarchy Parent: CVCL_C865 (v6.5)
